All three forms of Tumblestone currently generate in Mountains, which I like. However, I also feel that regular Tumblestone should spawn in Nether Wastes, Sky Tumblestone should spawn in Soul Sand Valley, and Black Tumblestone should spawn in Basalt Delta.
Here is what these forms of Tumblestone look like in those biomes:

At the moment, there isn't much reason to go to the Nether in Pixelmon. I have made a few suggestions to make the Nether more appealing to Pixelmon players, and this is one of them.
Soul Sand Valley's blue flames go well with the Sky Tumblestone, and the basalt and blackstone of the Basalt Delta match with the Black Tumblestone. Those forms of Tumblestone should spawn in those biomes. As for regular Tumblestone, it matches well enough with Nether Wastes. At the very least, Sky Tumblestone and Black Tumblestone should spawn in the biomes that I suggested.
